I have a client who is interested in hearing from others about how they manage data under these circumstances. They are staining with monoclonals and are gating on the "live" population. The percentage of monoclonal binding in this "live" population is relatively constant. The percentage of cells in the live gate changes dramatically in reponse to the treatment they are looking at. They want to adjust the percentage of monoclonal binding in relationship to the percentage of cells in the "live" gate.
