Dear calcium flux specialists, I have been looking at the intracellular calcium changes on monocytes. The problem is that after the addition of a stimulus the base line increases but the response is not presented as the Gaussian curve (rise and fall). I do have a negative control in which the base line stays the same and I have a positive control (ionomycin) where the response goes up and then down. Has anyone seen this before?
