I dug out some old records of a former colleague who had optimized a filter configuration for 2-color analysis of propidium iodide (PI) and FITC on a Coulter Profile (which allows a maximum of 2 colors). Instead of the usual FITC/PE setup, he used: split: 560SPdi FL1: 530SP FL2: 610LP Given the better spectral separation achieved by this combination, probably much less color compensation would be required while giving equivalent or better fluorescence intensities. Are there any thoughts on this?
