Monocyte depletion

We deplete monocytes from total peripheral blood mononuclear cells(PBMC)
by by incubating them on polystyrene tissue culture flasks in complete RPMI
medium for 45 min at 37'C, at 2-3x10^6 cells/ml, 12-15 ml/one T75 tissue
culture flask or 100mm tissue culture dish. (Monocytes will stick to
polystyrene
and we harvest the non-adherent cells). You can transfer the non-adherent
cells
to new flasks/dished and deplete them for a further 45 min if you need
complete depletion. Works fine with bovine and mouse PBMC.

Good luck
