Has anyone had experience with fluo-3/AM calcium detection? We have an investigator that would like to do this on resting and activated Th1 and Th2 cells. I have never run this before and would like any input anyone has on machine set-up (FACScan) and time course of mobilization. The paper they gave me does not include wavelenghts for excitation or emisison for bound or free calcium, nor does it define how they collected the sample(i.e. fixed time vs. fixed events). As you can tell by my questions here I am a novice at calcium detection by flow so any help is appreciated.
