To All: We are trying our hands at the Isolex 300 system for collecting CD34 cells. Our reccoveries are not what we would like but they are within range of expected. The problem comes up in the analsis of the CD34 cells on the pre selection sample. The bone marrow we use shows a bimodal distribution for CD34 positive cells. This is commonly seen versus a PBSC which gives a single peak. The calculated recovery on the isolex is depends on this number. The bright CD34 cells range from about 40-60% of total CD34 cells. This varying percentage as to how you analyze can take a recovery from 42% to 15% . Does anyone have any experience in these bimodal populations and how to best account for them? Jim Houston
