Alternative method for mouse spleens. Use Falcon #2350 Cell Strainer--
Has a 70 um mesh nylon membrane, comes sterile in individual packages, comes
in cases of 50. You can get from a number of lab suppliers, I've bought them
from Daigger and Lab Source.
It is a nice little plastic basket that will fit in the well of a 6-well
plate. We push mice spleens through this mesh with the plunger from a sterile
1 or 3 ml syringe. You can hold the baskets easily with a small hemostat.
Then you can throw away the basket and the plunger. No cleanup.
We used this method for SCID-Hu-PBL mouse model in animals infected with HIV.
Cells were good for co-culture, as well as flow cytometry.
