From: Robert C. Leif, Ph.D., Vice President & Research Director Newport Instruments To: cyto-inbox Subject: Position Available Job Information: NCI has funded for two years (starting 1 August, 1999) Newport Instruments’ STTR Phase 2 grant, Detection of Macromolecules with Quantum Dyes™. Newport Instruments is actively seeking a new full-time staff member to work on development of cytochemical procedures to convert Phoenix Flow Systems(www.phnxflow.com)flow cytometry technology for simultaneous measurements of cellular DNA, DNA synthesis, and apoptosis into a kit for microscopy. A Bachelor’s or Master's degree and experience in Cytochemistry and/or protein purification would be helpful. Please, contact Robert C. Leif, Ph.D, Vice President & Research Director Newport Instruments 5648 Toyon Road, San Diego, CA 92115-1022; Tel. & Fax (619) 582-0437; e-mail rleif@rleif.com. Background: Professor Vallarino of Virginia Commonwealth University and Dr. Leif (The PI) will continue their development of Quantum Dyes™, which are luminescent macrocycles with a central lanthanide ion. The europium containing Quantum Dye™ is commercially available from Research Organics, Cleveland Ohio (www.resorg.com). Previous work by Vallarino and Leif has shown that the europium(III) Quantum Dye™ is capable of covalent attachment to proteins and other macromolecules. The long lifetimes and narrow band emissions of lanthanides permit background free, time-gated luminescence measurements. The luminescence intensity of the Eu-macrocycle has been enhanced by the addition of other lanthanide ions to the solution, which results in the transfer of excitation energy to the Eu-macrocycle, producing a 10 to 100-fold increase in emission. This increase in emission in many cases eliminates the need for the special time-gated instrumentation. A. J. Bromm Jr., R. C. Leif, J. R. Quagliano, and L. M.
