I have been trying to find Fas ligand on lymphocytes from the intestinal mucosa and various cell lines. I have tried several antibodies and varied the staining conditions, but so far have had little success. I tried permeabilizing with saponin, inhibiting cleavage with a MMP inhibitor, and other things. My question: do you know of a good positive control that I can use to validate my protocols--possibly a readily available cell line with constituitive expression, or a method of induction on e.g. PBLs? I would appreciate any suggestions.
