Cindy asks:

Does anyone have experience using CD138 for detection of plasma
cells? What
kind of gating strategies do you use? What antibody combinations do
you
use?

Thanks,

Hi Cindy,

We have used CD138 from Serotec. Appears to work well, currently we
use 10 uL of neat antibody to 100 uL cell suspension. The antibody is
bright and shows positivity when CD19 maybe negative. It is used also
with a permeablizing/fixation product to show cytoplasmic expression of
both light and heavy chains. We tried this antibody on a few B-CLL and
NHL cases and demonstrated that the CD138 antibody to be negative.

Hope this helps,
Paul
