Hello, Flow Users:

Bone marrow megakaryocytes, the precursors of platelets, mature via
endoreduplication, forming normal populations of 4N-64N cells that become
larger as the nucleus expands with DNA.  These cells can be identified in
bone marrow preparations by gating on FSC (larger size) and FL1 after
staining for one of their surface glycoproteins, such as GPIIbIIIa, with a
FITC antibody.  Once gated, their ploidy content can be measured as a series
of DNA peaks by also staining with PI.
