HI FLOWERS I have a student that want work with reticulated platelets. we have see some protocols. In CURRENT PROCOLS IN CYTOMETRY, the author use a protocol with RNAase to test de fluorescence of Thiazol orange. To do this is necesary a permeabilization ( the TO binds to RNA and not need permeabilization).Some body have experience with permeabilization protocols for platelets ?. In the cytometry (CCC), 1998 (I dont remember the number) the author said that use a permeabilization buffer with HEPES, NaCl, Triton X-100, EDTA and " Glycerol 50 %". I think that is to much. What do you think ?
