Hello group, Anyone have advice on methods/enzyme cocktails useful for effectively dissociating cells from mouse stomach tissue? By effective, I mean so that we can collect representative cell types that maintain a high (or at least reasonably high) viability. We want to be able to detect surface expressed proteins, evaluate infiltrating leukocytes, and analysis of the lysosomal proton gradient (AO). We are presently using the medi machine. This machine is very effective at dissociating many types of tissue, but with stomach the viability is usually much lower that 50% (sometimes as low as 10%). Any ideas will be greatly appreciated.
