I think that this can be done. If you define "regions" on your histogram, most packages will store this information back to the histogram data file (usually much smaller file size than the LMD files) in the form of "Keywords" and their associated values. Then to extract this numerical data, use one of the "Purdue" utilities and your data will be put in a flat file for printing or placement into a data base. This has worked successfully for us. You can find access to these utilities on the U MASS cytometry page at: http://www.bio.umass.edu/mcbfacs/flowcat.html Also, you might email Steve Kelly (kelley@flowcyt.cyto.purdue.edu) for specific questions on these excellent utility programs. Phil Marder Lilly Research ----------------------------------------------------------------------------- reply to: Dear Group, I was wondering if there is a way to save statistical information from a histogram, such as % positive of a gated region, without saving a listmode of the sample. We would like to do this in order to facilitate running a large number of samples with pre-defined parameters, without generating the large FCS data files associated with those samples. We are currently running Cyclops software from Cytomation, with a Cicero Box attached to a FACScan, but would be interested if this can be accomplished with any other system, including CELLQuest and Beckman-Coulter's current version of software for the XL.
