Hi Mathi, I think if you look for any flow cytometer manual from Becton Dickson they have some statements to define a cell populations as lymphocytes. Even the CDC guide for enumerating CD4+ cell in HIV positive subjects have some information to monitor the gate of lymphocytes as far as gate purity and % of lymphocytes within the gate. Most of the procedures used the leucogate preps from any manufacturer. This reagent uses CD45 and CD14 in order to identify lymphocytes as bright CD45 and negative for CD14 in PBL from whole blood preparations. Of course if you are using PBMC you just will not have the presence of granulocytes, but lymphocytes still will keep their features as far as staining with CD45 and CD14.
