I have used the one suggested by BD from their lab methods book. The only thing I would advice you is if you will work with not fixed platelets it is good to use PBS suplemmented with EDTA 0,009-0,0013M to avoid platelet activation. I have a FACScalibur and the acquisiton is made using log scale for both FSC and SSC. I have no experience with coulter. Should be something similar, doesn't it?
