Hello to all, One of the PI's here at the University of Miami has a question. Here is what she wants to do: Immunize a mouse with an antigen, wait 10-14 days, harvest lymph nodes/spleen, do frozen sections, stain those frozen sections with antibodies specific for mouse IL-4 or IFN-gamma, and detect cytokine producing cells by immunohistochemistry or fluorescent microscopy. Has this been done? Actually, she wants to use confocal but doesn't think many people have access to confocal so they might have done it by conventional fluorescent microscopy. Any help would be appreciated.
