hi all, i've been attempting anti-PCNA staining with coulter mAb19F4 FITC labelled in combination with PE and PerCP Abs (CD33 or 38 and CD34). samples were analyzed on a FACScan. i have tried the fix and perm reagent protocol provided by coulter and caltag's fix and perm kit. results are less than spectacular on my target cells, cultured CD34+ human cord blood cells. channel seperation of PCNA pos./neg. (high/low?) populations is poor at best. are there any tricks to doing this or is this the best i can hope for? thanks in advance for your help. -brian white human genome sciences rockville, md brian_white@hgsi.com 301-309-8504 x2409
