high speed

Hello Dominique,
There is a considerable learning curve on the BD turbo sort option and running
a sort at high speed is definitely more demanding than at low speed. It takes
quite some time (considerably more than a month) and practice to reach a
really relaxed comfort level with the upgrade. The main thing is to get that
drop delay to be stable -- without a stable drop delay your purity and recovery
will always be terrible. Tricks that worked for us included moving the
sterilizing filter closer to the flow cell and installing a fan to keep the
electronic boards cool (we have a FACStar, not a Vantage). Glenn Paradis at
MIT has compiled a list of TSO users -- perhaps he will let you have the list
so that you can get their advice on specific questions. The BD option for high
speed sorting definitely works and is definitely worth the time and frustration
of getting used to the system.
