Hi- I have recently started a job which requires that I know something about DNA/ploidy analysis. 1) I'm wondering how others would report the example below 2) and am asking for opinions on the best published paraffin DNA procedural document(s). Ok, colon, formalin fixed, paraffin embedded, sent for DNA. Using Modfit, dipoid model, I get G0/G1 at 80 and a G2M at 167, I forget the exact numbers. But it wasn't too near 2x and my CEN's looked great - as linearity goes. With an aneuploid (1) model, I get G0/G1 at 80, G2M at 160, and an aneuploid peak of only 3.4%, too low of a percentage to call the population. The question is, which set of numbers do I use for reporting the diploid S-phase? It looked to me that the aneuploid model gave the more accurate (?) diploid figures, even though I couldn't call it aneuploid. Thanks in advance for any and all enlightenment.
