Hi! We have been using K/S analysis to determine kappa vs lambda expression on B-CLL cells which are well known to have an extremely weak expression. The system has to be standardized very well and one has to be sure that the fluorescence intensity on normal cells will not differ significantly between kappa and lambda. Then K/S analysis can be of help. Otherwise I agree that in NOT thoroughly standardized systems everything will be significantly different!
