Even if BDIS and PharMingen are very interested to sell their PerCP to dual laser FACSCalibur users, significant compensation is required between PerCP and APC on this instrument. However, the compensation is different for PerCP and APC when compared to PE-Cy5 and APC which can be used as well. PE-Cy5 conjugates in general have the significant advantage of being far brighter than PerCP conjugates. The only problem with PE-Cy5 is that some manufactures have problems in providing reagents with a lot-to-lot stable ratio of the PE and Cy5 emission. Furthermore, reagents from some but not all manufacturers tend to stick non-specifically to monocytes.
