Adipocytes and apoptosis

Hi folks,

I have a user who wants to examine apoptosis in porcine adipocytes and
pre-adipocytes.  The scattergrams of the initial preps looked horrible,
so I didn't think we had any intact cells.  They insist that the
cells are in good shape microscopically, but there is a 50-fold
variation in cell size.  Ok, I changed to log FS, but they don't
look much better.  Does anyone have any experience with adipocytes in
flow--specifically tips for preparation?  Is anyone looking at apoptosis
in these cells?  And finally (since _they_ think the results are
promising)--has anyone has any success with higher concentrations of
Triton X-100 in the tdt assay?  The B-M kit suggests 0.1%, and they have
tried up to 0.5%, but they were wondering if they could go higher.  I
suspect that we have a soup of debris which will only get soupier, but I
hate to pour water on their fires of enthusiasm.
