To All: Way back when I was taught that the jet velocity out of a nozzle for sorting was optimal at 10m/s.This is a jet in air system,no quartz flow cell.The method to calculate this jet velocity from volume equations for a cylinder are given in Howard Shapiros Practical Flow Cytometry.While taking these measurements may be crude,the overall approximation is good.What I would like to know is,Does anyone check the velocity out of the nozzle?If so, what do you set your sheath pressure at to get this velocity?What is the velocity you use for your system,either BD, COULTER, CYTOMATION, OR HOMEGROWN. I am talking the standard type sorters.Not your TSO from BD OR OTHER HIGH SPEED SYSTEM.
