Would anyone care to recommend fluorochrome-labelled anti-human Ig antibodies for detecting cell/platelet bound antibodies, and (preferably) distinguishing specific from non-specific binding. I have started to tinker with some anti-IgG-Fc (Fab) - but am experiencing horrific backgrounds at first attempts - so any tips/tricks on blocking, pre- or post -stain fix, whether whole blood techniques are common, and if so - when to lyse. What are people using in detection of leucocyte or platelet antibodies, flow crossmatches, etc? Robin Barclay Blood Transfusion Service Edinburgh
