I am attempting to lable Macrophages for identification with the flow. However, I have not been able to find any comercial markers. Are there any suggestions. We are looking at determining the % of Macs in a perotenial sample and from a spleen prep. in the deer mouse for use in a colormetric phagocytosis assay. I realize phagocytosis can be assessed on the flow, however, my advisor has chosen the colormetric relase of netural red, following phagocytosis. My main problem then, is that the # of Macs in each sample much be determined so that differences in OD readings are due to treatment effects and not the concentration of MAcs. Handcounts could give us % Macs, however, we will have 36 mice/day to assess this in while setting up a PFC assay, Mitogenic Proliferation assays, and running the phagocytosis assay and don't have time to preform hand counts on 36 samples. Therefore, the most cost effective and quickest method needs to be found. In the near future I will also need to assess phagocytosis and Mac% from fish Kidneys (Red Drum), Quail spleens, and Alligator spleens. Does anyone have any suggestions for a macrophage marker that would possibly work with deer mice, red drum, bobwhite quail, or alligators. Obviously something Mac specific but not species specific.
