Preparation

SBA has developed proprietary technology for the conjugation of antibodies to a variety of enzymes. All antibodies are conjugated under defined conditions to yield optimally labeled products with high specific activity and excellent stability.

Alkaline phosphatase (AP). Calf intestine alkaline phosphatase (specific activity = 3000 U/mg enzyme protein) is conjugated to antibodies using a heterobifunctional linking reagent under conditions to ensure that no unlabeled ligand remains in the conjugate.

Horseradish peroxidase (HRP). Horseradish peroxidase (specific activity = 1000 U/mg enzyme protein) conjugates are prepared using a modification of the periodate method. HRP-labeled reagents are produced to yield conjugates of optimal size and activity for ELISA, blotting and immunohistochemical applications.

B-galactosidase (BGAL). Affinity purified antibodies are labeled with B-galactosidase (BGAL, specific activity = 600 U/mg enzyme protein) using a heterobifunctional linking reagent. B-galactosidase-labeled reagents are useful for cellular ELISA where endogenous AP or HRP may lead to nonspecific background.

Working Dilution

The dilutions shown on the Table above are only recommendations. The optimal dilution for a particular application should be determined by the investigator.

Specificity

 To ensure lot-to-lot consistency, each enzyme-labeled antibody is tested by ELISA for conformance with characteristic of a standard reference reagent.

Form

AP conjugates are supplied as a 1.0 mL stock solution in 50% glycerol/50% 50 mM Tris, pH 8.0, containing 1mM MgCl2 and 0.1% NaN3 as preservative.

HRP conjugates are supplied as a 1.0 mL stock solution in 50% glycerol/50% phosphate buffered saline, pH 7.4. No preservative added.

BGAL conjugates are supplied as a 1.0 mL stock solution in 50% glycerol/50% phosphate buffered saline, pH 7.4, containing 0.1% NaN3 as preservative.

Storage and Stability

Enzyme-labeled antibodies can be stored for several months at 2-8°C without appreciable loss of working titer. For long-term storage of the conjugates, the labeled antibodies should be kept at -20°C.
