diagnostic or in vivo use.
Specificity
This antibody recognises the CD14 antigen (LPS receptor) expressed strongly on the surface of monocytes, weakly on the surface of granulocytes, macrophages, dendritic cells and B-cells. On flow cytometry it stains >90% of human peripheral blood monocytes.
The antibody is LPS neutralising.
Performance
Flow Cytometry:
Recommended use of FITC, PE and APC-conjugates:
Whole blood: 100mL blood + 20 mL mAb
Cell preparations: 50mL (500 000) cells + 20 mL mAb
Wash is recommended to yield optimal results. For more details, see Flow cytometry methods page

Storage
Unconjugated antibody: The antibody is diluted in phosphate buffered saline, pH 7.4.
For long term storage, -20ºC is recommended, but it may be stored for short
term use at 4ºC. The expiry date on the vial refers to storage at -20ºC.
Repeated freeze/thawing is not recommended.
Conjugated antibody: The antibody is diluted in phosphate buffered saline, pH 7.4 containing 0.2% BSA.
Storage at 4ºC. The expiry date on the vial refers to storage at 4ºC.
Preservative
Sodium azide is used as a preservative at a concentration of 0.1%. Sodium azide under acid conditions yields hydrazoic acid, an extremely toxic compound.
Antigen Structure and function
1
CD14 is a GPI-linked glycoprotein.
CD14 is a receptor for LPS (Lipopolysaccaride complex) and for LPS-binding protein (LBS).
Antigen tissue distribution
1
The CD14 antigen is expressed mainly on cells of the myelomonocytic lineage including monocytes, macrophages and Langerhans cells.
It is also expressed at low levels on neuthrophils and on human B-cells.
