Go vs. G1 discrimination

The probe which supravitally may distinguish Go from G1 cells is
rhodamine 123. We have noticed that uptake of this fluorochrome by
quiescent Go lymphocytes is several times lower compared to
mitogenically stimulated cells (Proc. Natl Acad. Sci. USA, 78: 2383,
1981). The increase was observed during G0 to G1 transition and then
during cell progression through S and G2. However, in light of the
evidence that rhodamine 123 is pumped out very rapidly from stem
cells by P glycoprotein, it may happen that one has to discriminate
between very weak labeled cells (Go) and weak labeled ones
(G1/S/G2M). Another parameters to consider is forward light scatter,
pulse width or Coulter volume. Go cells are smaller compared to G1
cells, and therefore generate lower intensity forward light scatter
signal (PNAS, as above).
