It seems CNFDA was available from molecular probes a few years ago
but it is listed as "to be discontinued when our current stocks are
depleted" in ed 5 (1992-4) of the handbook - and not listed at all in
the 1995/6 UK price list.
Others mentioned TO-PRO-3, (and TO-TO-3). I've tried TO-PRO-3 with
yeast and the results are certainly believable. I am a bit concerned
about stories that it may enter viable cells too. I analysed my
samples immediately after adding the dye and didn't detect a problem
with staining of viable cells - although in the flow world it isn't
always easy to analyse immediately ;-)
Another suggestion was carboxy SNARF-1 AM acetate ("a red excitable
pH indicator that is only cleaved to fluorescent SNARF by live
cells"). I haven't followed this lead any further yet.
DiTBAC4(5) was mentioned here on the list.
More than half of the responses were asking for this information so I
would assume that there is a lot of interest in red excitable dyes
(hint to any dye manufacturers out there!!!).
