No...DAPI is not a vital stain. Some while back someone told me it was
and we tried it (compared it with H33342). No DNA histogram. I haven't
looked at my Mol. Probes guide yet, but I think it reads as DAPI being
perhaps a "viability" probe -- like PI -- and only dead cells will label.
Whatever....bottom line...you have to fix your cells. If you do, however,
you will find DAPI out-performs H33342 significantly.
