I intend to use fluorogenic beta-galactosidase substrate to study gene
expression in SW480-cells that are stable transfected with the gene encoding
beta-gal (reporter-gene). I will analyse beta-gal activity in conjunction
with other cell markers. If it is possible with these substrates, I also
want to sort high and low expressing cells and do a further study of
membrane receptors and intracellular components.
According to Molecular Probes there are various sustrates available: FDG,
carboxy-FDG, dichloro-FDG, chloromethyl-FDG(DetectaGene) and
C12-FDG(ImaGene). But I am not familiar with the use of these.
If you have any experience with these or other beta-gal substrates used in a
similar manner, I will find it very beneficial to have comments, hints,
references etc.
