The specificity of staining by monoclonal antibodies to mouse CD antigens should be verified by establishing the non-specific reagent binding to the target cell population. It is recommended that a concentration and isotype matched mouse myeloma immunoglobulin be included as a negative control for each monoclonal antibody reagent used in the immunoassay procedure.

Sigma Mouse Isotype Controls are mouse myeloma proteins tested for use as controls in flow cytometry.

Sigma Mouse Isotype Controls are supplied to match the concentration of Sigma purified and/or conjugated monoclonal antibodies to CD antigens. These products are liquid, 100 mg/vial, in 0.5 ml of 0.01 M phosphate buffered saline (PBS), pH 7.4, containing 1% BSA and 0.1% sodium azide as preservative. DUAL-TAG controls contain 100 mg of each conjugate, or 200 mg/vial in 1 ml.

Mouse Isotype Controls conjugated to FITC, R-Phycoerythrin (PE) or Quantum RedTM may be used as controls for monoclonal antibodies to CD antigens in direct immunofluorescence procedures. Each isotype control is prepared with a F:P (fluorochrome:protein) molar ratio in the range similar to the specific monoclonal antibody conjugates offered by Sigma.
