CytoStain™ Cell Fixation and Permeabilization Kits

This kit enables the one step fixation and permeabilization of cells necessary prior to the staining of intracellular cytokines with fluorochrome-conjugated anti-cytokine antibodies. This kit provides two reagents: Cytofix/Cytoperm™ fixation/permeabilization solution and Perm/Wash™ buffer. After fixation/permeabilization using the Cytofix/Cytoperm™ solution, the Perm/Wash™ buffer is used to wash the cells and to dilute the anti-cytokine antibodies for staining. It is important that the Perm/Wash™ buffer be used for dilution of anti-cytokine antibodies, rather than a standard staining buffer, in order to maintain cells in a permeabilized state for intracellular staining.

In addition to the fixation/permeabilization and diluent/wash solutions included in the Cytofix/Cytoperm™ kit, the Cytofix/Cytoperm Plus™ kit provides GolgiStop™, a protein transport inhibitor for use during the last few hours of cell culture. Addition of GolgiStop™ to cell activation cultures will block intracellular transport processes, thereby resulting in the accumulation of most cytokine proteins in the Golgi complex and enhancing cytokine staining signal.
