PSGL-1 displays specific oligosaccharide sequences which interact with the lectin domain of P-selectin. The epitope on PSGL-1 recognized by the PL1 antibody is in close proximity to this lectin binding site and the antibody completely inhibits the binding of P-selectin to PSGL-1 in a variety of assays. In contrast, the PL2 antibody does not block any binding ofP-selectin to PSGL-1. PL1 and PL2 antibodies react with all leucocytes including circulating dendritic cells, but not with platelets and erythrocytes. Both antibodies can be used in flow cytometry and in immunohistochemistry on frozen and paraffin embedded tissue sections.
