Laboratory Profile

Director: Dr. Sergio Lucretti

1.Facilities available: 200 m 2 facility containing instrumentation lab, molecualr and cellular biology labs, plant tissue culture facilities, growth chambers,

2. Instrumentation:

Flow cytometry: Becton Dickinson FACStar PLUS dual laser (argon 5W and 2W visible); also available Partec PasII(Hg lamp flow-cytometer).

Other major items: Microspectrofluorimeter Leltz-SP for analysis of light absorption and fluorescent emission from microscopic specimens; normal and inverted microscopes equiped with fluorescence; also available a BioRad MCR 600 confocal microscope.

Computer and Analytical: Al flow-cytometric data are compatible with MS-Dos and HP Pascal 3.2-based instruments; non cytometric software expertise on MacIntosh computers.

3. Specialization areas: Flow-karyotyping and sorting of plant chromosomes; Mono- and biparametric cellcycle analysis of in vivo and in vitro explants; Plant protoplast isolation and sorting; Somatic hybridization of plant protoplasts; Direct mcroencaps ulation of sorted particles by flow cytometry.

4. Training Opportunities: Cell-cycle synchronization of organs and cell suspensions for chromosome and nuclei isolation; Plant cell-cycle analysis by BrdU and immunolabelling; In vitro plant tissue culture; Somatic hybridization of protoplasts and plant regeneration; Plant transformation. Our lab has extensive experience on DNA ploidy evaluation from different explant sources and sorting of protoplasts under biotic stress conditions.

5. Special courses offered: BD FACStar PLUS usage; Plant protoplast isolation, fusion and plant regeneration in Citrus,

6. Graduate Program: None.

7. Local Accomodations: In Rome, near ENEA headquarters, arrangement with hotels as low as 70 $/single room.

8. Transportation: Local airport is Rome International Airport (Leonardo da Vinci). Several free shuttle bus connections from ENEA headquarters to C.R. Cassacia - research center 30 km north-west of Rome.

9. Cost of Internships: base rate of about 400 $ per week will cover most of laboratory cost and flow cytometry, depending on what studies are required. The period for internships must be arranged directly.
