Hello,
This is a brief announcement of a utility that I have written to aid
conversion of and data extraction from FCS Format files.
It goes by the name of FCS Assistant, and is available from Eric Martz's
Software Catalogue (http://www.bio.umass.edu/mcbfacs/flowcat.html) and
ftp server (marlin.bio.umass.edu/pub/flowcyt/fcs-assistant) and my own
web site and ftp server (facsmac.med.cam.ac.uk).
It reads FCS1.0 and FCS2.0 file formats and saves FCS2.0 as well as
exporting text files. It can handle up to 16 parameters, and can be used
to remove superfluous parameters.
It will calculate the ratio of two parameters, and add an estimate of time
(based on sample duration and the number of events), it will also
calculate the mean of one parameter (eg Indo-1 ratio) versus another (eg
time) for plotting kinetic data (export only).
The three text file formats it outputs are:
.LGD- Line graph data for kinetic plots
.HD- Histogram data for histogram plots
.RTT- Raw tab text for spreadsheet analysis of list mode data.
It will open files from any platform (swapping bytes if necessary), and
converts them into a minimal FCS2.0 format that is compatible with Cellquest,
I will try to get it to save LYSYS and earlier formats if there is a demand.
If you have problems getting the program please let me know and I'll try
to help, it's easiest if you use Fetch or Netscape with binhex
translation switched on to get the .hqx version the .sea version must be
got using macbinary mode.
