Indo loading

Hello

I was wondering if anyone has experience loading COS-7 cells with
Indo for calcium mobilization analysis. We've been having a tough
time with this. The Indo seems to be ok, since we've loaded other cells
nicely with the same stock. We are using the standard protocols for
loading and have varied time from 1/2 to 1 hour, also varied the Indo
conc., also tried loading the cells 1/2 hour after they've been taken off
the dish (to let them calm down ).
We haven't tried the Pluronic trick yet, and haven't tried loading in
the flask before lifting the cells. Any suggestions.
