Because most instruments flush their sample line with the sheath fluid,
the sample line is filled with the sheath buffer when a new sample is
started. We have seen not only changes in fluorescent levels but also light
scatter when using DI for sheath fluid. Once enough sample is run to unflush
the line, light scatter and fluorescence return to expected values. DI may
work fine with fixed samples, but care should be taken when using it for all
samples.
