Here at Dartmouth we did a comparison of distilled water and PBS as sheath
fluid a few years ago. Percent positive seemed to be unaffected but level
of fluorescence (MFI) was reduced and variable for distilled water. In fact,
our comparison was prompted by variations in staining intensity during the
time we used distilled water. We checked our distilled water source and
discovered slight pH variations from day to day. When water pH went down so
did our fluorescence levels. While there is not supposed to be any mixing of
sheath and sample, our tests made us think that some interaction must occur.
We decided to switch our sheath to PBS pH 7.4 and have had no problems since.
