We want to run indo-1 and cell surface markers simultaneously on an EPICS
753. We could run FITC and PE along with the indo but that involves a
pulling the dye laser off the optical bench. We could also install the
optics from an EPICS 752 to steer the dye laser pump beam around the dye
head, but there is still a major hassle involved in realigning the pump
beam with the dye head once the 752 optics are pulled and the Z mirror
is reinstalled between the pump beam and the dye head. Several years
ago I tried using indo plus Texas Red exciting with UV and 600 nm from
the dye laser. The Texas Red signal was compromised I suspect from a
suboptimal optical filter setup. It is obviously much easier to
configure the instrument with the Dye head left on the optical bench.
If anyone has run indo-1 and some other fluorochrome excited with a
dye laser or HeNe (Texas Red, APC, Cy5), I would be interested to know
what optical filter setup you used.
