PC Software Upgraded (MFI)

Version 3.4 of my free PC-based list-mode cytometry data analysis program
MFI.EXE is now available for retrieval by anonymous FTP from
flowcyt.cyto.purdue.edu in directory pub/martz. For detailed instructions on
use of FTP, email a request to me.

The MFI.EXE program includes extensive help including a tutorial. MFI
excels at printing a compact table of corrected median fluorescence
intensities from a group of list mode files in batch runs (4 sec/file).
Each run is organized by tagging filenames on a master list, including gate
assignment (9 gates are remembered simultaneously), control marking, and
filename sequencing. Dot plots (used for graphic gate setting) and
optionally smoothed histograms (optionally overlayed) can be viewed and
printed. MFI works with files from most B-D and Coulter cytometers. Gated
ASCII list mode and histogram data files can be exported for use in generic
plotting software.

In MFI 3.4, the handling of pileups of off-scale events at the ends of
histograms has been redesigned; it is now more consistent, flexible, and
less likely to lead to misinterpretation. Constants for conversion of
fluorescence intensities to KMESF (kilo molecules of equivalent soluble
fluorochrome) can now be entered separately for each fluorescence channel.
List-mode data can now be written, optionally gated, into files in ASCII
format for import into generic scientific plotting software. Several bugs
were fixed. Details will be found in the file MFI34.DOC available by
anonymous FTP as above.

Users of previous versions of MFI (3.3f being the most recent) please note
that MFI 3.4 will not recognize configurations set-up with MFI 3.3. Thus,
you may wish to rename your old MFI.EXE to MFI33.EXE so it will remain
available for use with old MFI.CFG files. The old version will remain
available by anonymous FTP as MFI33F.EXE should you wish to retrieve it.

MFI 3.4 requires about 530K free memory for average use, up 15K from MFI
3.3f. I will be working to reduce the memory required for operation of MFI
in the next release.

model. I would especially like to know whether, and for what purposes, you
find MFI advantageous vs. your alternative software, and what alternative
software you have compared it to. Future enhancements depend on user
feedback.
