Bird blood and lizard blood

Does anyone have any experience running bird or lizard blood for
DNA content and/or sex determinations? We have been trying to use
a protocol with 0.1% sodium citrate + 0.1% Triton X-100 as the buffer
and staining with P.I. We have been having problems with the stability
of the fluorescence signal as the samples are being run. Any suggestions
or hints would be helpful.
