CD4 enumeration

This query is directed primarily to clinical users, but ALL opinions are
welcome.
Given the numerous literature citations, and current dogma, regarding the
absolute necessity of maintaining specimens for CD4 enumeration at ambient
temperature:

1) Is there documentation as to what actually happens to the CD4 receptor
and/or the cell(s) bearing these receptors, once the specimen is subject to
refrigeration?

2) What is the validity, reliability, relevance, and/or experience in
enumerating CD4 on peripheral blood mononuclear cells that have been frozen
and thawed using standard tissue culture techniques, ie, controlled rate
freezing, liquid nitrogen storage, etc.

3) If this has been successfully performed, are literature citations
available?

Apologies if a) this subject is taboo, and/or 2) it has been dealt with
previously in this forum.

Thanks in advance,

Marc Langweiler
