Hi,
We are trying to use BrdU staining to identify cycling cells in 32D
cultures (murine myeloid precursors). Control asynchronous cells in
log phase showed no positive cells with anti-BrdU. PI stain showed
plenty of S-phase cells. Control 3T3 cells stained as expected with
anti-BrdU. We use 10 micromolar BrdU for 1 hr before fixing. Is there
something unusual about 32D cells with respect to this protocol? Any
ideas would be appreciated.
