We are using PKH2-GL (green fluorescent) and PKH26-GL (red-orange
fluorescent) at 1 =B5M and 2.5 =B5M, respectively and 10e7 cells/ml=
(staining: 5
minutes at room temperature) for membrane labelling of different (tumor)
cell lines and subsequent analysis of antibody dependent effector/target
cell aggregation. Labelling conditions may vary and have to be optimized for
special experimental requirements and cell lines.
FACS analysis (FACScan) is performed using 488nm excitation wave length and
530 and 585 nm emission filters. Both labels are stable for at least several
days. Kits are available from Sigma.=20
