Re: retic analysis with high WBC

Andrea - I would like to modify your statement regarding WBC interference
with FCM retic analysis withg thiazole orange. WBCs can potentially
interfere with the retic % and immature reticulocyte fraction (IRF) only
if one does not gate out the WBC based upon there higher TO fluorescence
(approx 5-10X that of retics). See Cytometry 14:318-26, 1993, Heme/Onc
Clinics of N Amer 8:617, 1994, cytometry (CCC) 22:35-39, 1995, or
Laboratroy Hematology 2:2-8, 1996 for reviews. I highly recommend Verity
Software's ReticFit program, which simplifies FCM retic data analysis
from any instrument with regard to both elimination of WBCs and IRF
