DNA staining of fish muscle nuclei

Hi,
I'm seeking input from any people who happen to have experience in
staining DNA in dissociated nuclei of fish muscle cells. Searching so far
has revealed only one paper using a DAPI staining method, however if someone
out there has experience using PI or other probes in this area I'd
appreciate hearing from you.
Also, in order to keep the dissociated nuclei from clumping the
student involved in this project has been adding Ca++ to his medium
(balanced salt solution). Does anyone know if this will interfer with PI
staining ?
