Subject: Re: FISH by flow

There have been several attempts to perform FISH by flow. One was published
by a japanese group last year. We have also attempted it. None of the
published or published efforts has been very successful.

The primary problem is the relatively small amount of fluorescence of the
typical FISH hybridization. Flow cytometers require a fairly high amount of
fluorescence intensity on the targets they analyze. FISH probes rarely have
this level of intensity (with the possible exception of "paint" probes).

A secon problem is the cost of the probes themselves. The hybridization must
be performed in suspension, not on a slide, this vastly increases the probe
cost.

I hope this response is suitable. If the physician who made the inquiry has
any other questions I would be glad to answer them.
