NK Activity by Flow

Hello Out There!
I am trying to determine NK activity using flow cytometry by
staining my K562 targets with Calcein AM, incubating with effectors
and then staining all cells with PI. Dead targets should be the only
dual positives. Does anyone out there has any suggestions or comments
on this technique. Any input will be gratefully accepted.
